Don't know if you have fixed this issue or not but from the outside looking in, I would ask, Is it always the same cylinder missing or does it move. Same cylinder would indicate things like a valve sticking or a lifter collapsing or not pumping up. Maybe the new tuneup parts just mask the issue..
